Olvera Street is the birthplace of the City of Los Angeles, otherwise known as El Pueblo Historic Monument. The colorful village features 27 historic buildings with a traditional Mexican style plaza area. Wander around the marketplace and shop for souvenirs including handcrafted Mexican wares typical of old Mexico. There are also free docent tours by Las Angelitas.

This is the top level of the Falkirk Wheel, Falkirk, Scotland - it is the worlds first and only rotating boat lift and carries canal boats from the lower level of one canal to the upper level of another canal. This is the Bahá'à House of Worship on Lake Michigan located in Wilmette, Illinois. There were six original Temples constructed (One on each continent) and this is the only one in the Western Hemisphere. Photo was taken from my friends front yard this morning. http://en.wikipedia.org/wiki/Bahá'Ã_House_of_Worship
Forgot to add: The temples are all different so this is the only 'one' anywhere architecturally speaking.

I live in San Francisco and one afternoon while drinking beer at the Zeitgeist a whole crowd of naked bikers stumbled in and proceeded to drink in the beer garden with nothing but helmets and shoes on...for hours! I got this shot of three of them leaving and minutes later they were in a drunken pile of naked flesh and bikes after wiping out and bringing one another down :(

There are a lot of things that are unique to this city, but this one happens to be (briefly) visible on my morning commute. She's hard to see in this picture, but she's certainly one-of-a-kind.

"The old town" is a small town in the middle of the city Aarhus, made out of old buildings scheduled as monuments, imported from all over Denmark. It is a "Live" museum, and - i think - unique for Aarhus, Denmark.
